Agouti overexpression in a transgenic model regulates integrity, permeability and electrogenic amino acid transport in zebrafish intestine

Overexpression of asip1 in transgenic zebrafish disrupts dorsoventral pigment pattern addition to increasing food intake levels and linear growth. A higher feed is unnecessary fish enable larger heavier plausible explanation may rely on the enhanced feeding efficiency mediated by improved nutrient absorption animals. To test this hypothesis, wide scope transcriptomic techniques were used elucidate potential pathways involved intestinal epithelium permeability/integrity. In addition, electrogenic capacity for amino acid transport was analysed. Transcriptomic analysis reveal that acid, monocarboxylates, ionic vitamin transmembrane transporters substantially modified. Enrichment also revealed an inhibition lipid metabolism down-regulation KEGG related membrane integrity suggesting augmented laxity enhance paracellular transport. Electrophysiological experiments carried out Ussing chambers show overexpression decrease membraned tissue resistance (Rt), indicating a modification barrier function ASIP1 Similarly, permeability zebrafish. Both Rt increase point ASIP1-dependent function. Electrogenic animals providing strong indication can extract more acids from their diet at similar levels. electrophysiological results suggest asip1-overexpressing display extension which explains growth absence intake. The potentially uptake suggests melanocortin system, specifically overexpression, target development genetically engineered displaying performance no differential accumulation.
